I don't know if it is so possible in one day, except if you leave from Budapest too early in the morning.
Probably you plan to pass through Serbia and then Bulgaria to Greece, don't you?
The border between Bulgaria and Greece is now open as EU states, but I think you will loose some time from Hungary to Serbia and then to entry to Bulgaria...
You have to calculate this on your time planning.
I haven't done this road...and I don't have a personal experience... :(

From the border of Bulgaria the distance to Serres is about 40/45km.
There is a "quicker" way (indicated to www.viamichelin.com ) passing through Skopia, but when you will entry to Greece you have to go to Thessaloniki and then to Serres which is longer.
